amfiSure HiFi High Fidelity DNA Polymerase

amfiSure HiFi High Fidelity DNA Polymerase combines GenDEPOT’s Taq DNA Polymerase with a high fidelity, proofreading polymerase. This enzyme blend has the 5'-3' exonuclease activity of Taq DNA Polymerase as well as the 3'-5' exonuclease activity of the proofreading enzyme. amfiSure HiFi High Fidelity DNA Polymerase increases fidelity up to 6-fold over Taq DNA Polymerase alone and allows for amplification of longer products. Although a large range of PCR product sizes can be generated from a variety of DNA templates, optimization may be required for targets from complex templates. Additives such as betaine, trehalose, glycerol, or DMSO can be used to aid longer target amplification.

This enzyme blend generates PCR products whose ends are compatible with either blunt-end or TA cloning procedures, with A-tailed ends favored over blunt ends in an approximately 3 to 1 ratio. Thus, this product is useful in both blunt-end and TA cloning procedures and kits. Do not use dUTP as a substitute for dTTP in a nucleotide mix because the proofreading polymerase in the enzyme mix is inhibited by dU-containing DNA.
